    摘要:

    鳄雀鳝(Atractosteus spatula)作为中国《重点管理外来入侵物种名录》中的重点监管入侵物种,对本地水生生物多样性构成了严重威胁。为满足鳄雀鳝入侵早期阶段快速、准确检测的需求,基于环境DNA (environmental DNA, eDNA) 技术,开发了一套高特异性、高灵敏性的鳄雀鳝快速检测体系。通过线粒体基因组比对分析,设计出一组鳄雀鳝特异性重组酶聚合酶(Recombinase polymerase amplification, RPA)引物与探针。验证结果表明:(1)该引物探针组在组织样本、养殖水体样本以及中国水产科学研究院珠江水产研究所的养殖水体样本中均能稳定高效地扩增目标DNA。(2)特异性验证显示,该引物探针组仅对鳄雀鳝产生阳性信号,对包括眼斑雀鳝(Lepisosteus oculatus)在内的11种非目标鱼类无交叉反应。(3)灵敏性验证显示,该引物探针组的最低检测限可达15.88 copies/μL DNA,表现出较高的灵敏度。(4)该检测体系可在38 ℃条件下于15 min内完成RPA扩增,并结合侧流层析(Lateral flow, LF)试纸条在5 min内实现条带读取。本研究首次建立了基于环境DNA技术的鳄雀鳝RPA-LF快速检测体系,为水生生态系统中入侵物种的早期预警与科学管理提供了技术支撑。

    Abstract:

    Atractosteus spatulais (alligator gar), as a key regulated invasive species in China's Catalogue of Key Managed Invasive Alien Species, has posed a severe threat to local aquatic biodiversity. To meet the demand of rapid and accurate detection during the early stages of alligator gar invasion, this study developed a specific, highly sensitive and rapid detection system based on environmental DNA (eDNA) technology. A set of alligator gar -specific recombinase polymerase amplification (RPA) primers and probes was designed by mitochondrial genome alignment analysis. The validation results indicate: (1) The primer-probe set stably and efficiently amplified target DNA from tissue samples, aquaculture water samples, and aquaculture water samples from the Pearl River Fisheries Research Institute Chinese Academy of Fishery Sciences, Guangzhou, Guangdong, China. (2) Specificity validation demonstrated the primer-probe set produced positive signals exclusively for A. spatula is with no cross-reactivity to 11 non-target fish species, including Lepisosteus oculatus. (3) Sensitivity validation demonstrated the detection limit reached 15.88 copies/μL DNA for the primer-probe set. (4) RPA amplification can be completed within 15 min at 38 ℃ and the lines can be read within 5 min combined with lateral flow (LF) dipsticks. This study firstly established a rapid detection system for A. spatulais using RPA-LF based on eDNA, providing technical support for early warning and scientific management of invasive species in aquatic ecosystems.
